| Patent Claims: | 1. An isolated cell line comprising a recombinant vector encoding a single chain FVIII polypeptide, wherein the single chain FVIII polypeptide comprises residues 20 to 1457
of SEQ ID NO: 8.
2. The cell line of claim 1, wherein the single chain FVIII polypeptide further comprises a heterologous moiety. 3. A polynucleotide encoding the single chain FVIII polypeptide of claim 1. 4. A vector comprising the polynucleotide of claim 3. 5. The cell line of claim 1, wherein the cell line is a mammalian cell line. 6. The cell line of claim 2, wherein the heterologous moiety is a half-life extending moiety. 7. The cell line of claim 6, wherein the half-life extending moiety is an immunoglobulin constant region or a portion thereof, albumin, albumin binding polypeptide, PAS, the C-terminal peptide (CTP) of the .beta. subunit of human chorionic gonadotropin, polyethylene glycol (PEG), hydroxyethyl starch (HES), albumin-binding small molecules, or combinations thereof. 8. The cell line of claim 1, wherein the cell line further comprises a polynucleotide specific for a FVIII polypeptide processing enzyme. 9. The cell line of claim 1, wherein the single chain FVIII polypeptide further comprises an immunoglobulin constant region or a portion thereof. 10. The cell line of claim 1, wherein the single chain FVIII polypeptide further comprises albumin. 11. A polynucleotide encoding the single chain FVIII polypeptide and the half-life extending moiety of claim 6. 12. A vector comprising the polynucleotide of claim 11. 13. A polynucleotide encoding the single chain FVIII polypeptide and the immunoglobulin constant region or a portion thereof of claim 9. 14. A vector comprising the polynucleotide of claim 13. 15. An isolated cell line comprising a recombinant vector encoding the amino acid sequence as set forth in SEQ ID NO: 8. 16. A vector encoding the amino acid sequence as set forth in SEQ ID NO: 8. 17. A method for producing a single chain FVIII polypeptide comprising culturing the cell line of claim 1 under conditions sufficient for production of the single chain FVIII polypeptide. |
