Abstract:The present disclosure relates to certain (2S)—N-[(1S)-1-cyano-2-phenylethyl]-1,4-oxazepane-2-carboxamide compounds (including pharmaceutically acceptable salts thereof),that inhibit dipeptidyl peptidase 1 (DPP1) activity, to their utility in treating and/or preventing clinical conditions including respiratory diseases, such as asthma and chronic obstructive pulmonary disease (COPD), to their use in therapy, to pharmaceutical compositions containing them and to processes for preparing such compounds.

Patent Claims: 1. A pharmaceutical composition comprising a compound of formula (I), a pharmaceutically acceptable diluent, and a disintegrant, wherein the compound of formula (I) is present in the composition from 0.10 to 50% weight percent of the pharmaceutical composition, and wherein, R1 is R2 is hydrogen, F, Cl, Br, OSO2C1-3alkyl, or C1-3alkyl; R3 is hydrogen, F, Cl, Br, CN, CF3, SO2C1-3alkyl, CONH2 or SO2NR4R5, wherein R4 and R5 together with the nitrogen atom to which they are attached form a azetidine, pyrrolidine or piperidine ring; or R1 is X is O, S or CF2; Y is O or S; Q is CH or N; R6 is C1-3alkyl, wherein said C1-3alkyl is optionally substituted by 1, 2 or 3 F and optionally by one substituent selected from OH, OC1-3alkyl, N(C1-3alkyl)2, cyclopropyl, or tetrapydropyran; R7 is hydrogen, F, Cl or CH3; or a pharmaceutically-acceptable salt thereof.

The oral composition of claim 1, wherein R1 is X is selected from O, S or CF2; Y is selected from O or S; Q is selected from CH or N; R6 is C1-3alkyl, wherein said C1-3alkyl is optionally substituted by 1, 2 or 3 F and optionally by one substituent selected from OH, OC1-3alkyl, N(C1-3alkyl)2, cyclopropyl, or tetrahydropyran; and R7 is selected from hydrogen, F, Cl or CH3.

The pharmaceutical composition of claim 1, wherein, R1 is X is selected from O, S or CF2; Y is selected from O or S; R6 is C1-3alkyl, wherein said C1-3alkyl is optionally substituted by 1, 2 or 3 F and optionally by one substituent selected from OH, OC1-3alkyl, N(C1-3alkyl)2, cyclopropyl, or tetrahydropyran; and R7 is selected from hydrogen, F, Cl or CH3.

4. The pharmaceutical composition of claim 1, wherein, R1 is or a pharmaceutically acceptable salt thereof.

5. The pharmaceutical composition of claim 4, wherein, X is O; R6 is C1-3alkyl; and R7 is hydrogen; or a pharmaceutically acceptable salt thereof.

6. The pharmaceutical composition of claim 1, wherein, R1 is X is O; R6 is C1-3alkyl, wherein said C1-3alkyl is optionally substituted by 1, 2 or 3 F; and R7 is hydrogen.

7. The pharmaceutical composition of claim 1, wherein, R1 is X is O; R6 is C1-3alkyl; and R7 is hydrogen.

9. The pharmaceutical composition of claim 1 which is (2S)—N-{(1S)-1-cyano-2-[4-(3-methyl-2-oxo-2,3-dihydro-1,3-benzoxazol-5-yl)phenyl]ethyl}-1,4-oxazepane-2-carboxamide: or a pharmaceutically acceptable salt thereof.

10. The pharmaceutical composition of claim 1 which is (2S)—N-{(1S)-1-cyano-2-[4-(3-methyl-2-oxo-2,3-dihydro-1,3-benzoxazol-5-yl)phenyl]ethyl}-1,4-oxazepane-2-carboxamide:

11. A method for treating an obstructive disease of the airway, comprising orally administering to a patient in need thereof, the pharmaceutical composition of claim 1.

12. A method for treating an obstructive disease of the airway, comprising orally administering to a patient in need thereof, the pharmaceutical composition of claim 10.

13. The method of claim 11, wherein the obstructive disease of the airway is bronchiectasis.

14. The method of claim 11, wherein the obstructive disease of the airway is cystic fibrosis.

15. The method of claim 11, wherein the obstructive disease of the airway is asthma.

16. The method of claim 11, wherein the obstructive disease of the airway is chronic obstructive pulmonary disease (COPD).

17. The method of claim 12, wherein the obstructive disease of the airway is bronchiectasis.

18. The method of claim 12, wherein the obstructive disease of the airway is cystic fibrosis.

19. The method of claim 12, wherein the obstructive disease of the airway is asthma.

20. The method of claim 12, wherein the obstructive disease of the airway is chronic obstructive pulmonary disease (COPD).
