1. A pharmaceutical composition formulated for subcutaneous administration comprising a controlled-release CNP agonist comprising a CNP moiety (-D) having the amino acid sequence of any of SEQ ID NOS:1-95, wherein disulfide bonded cysteines at positions in the amino acid sequence of the CNP moiety corresponding to 22 and 38 of SEQ ID NO:24 enclose a ring moiety, the CNP moiety being conjugated to a polymer via a linker cleavable to provide controlled-release of the CNP moiety, wherein the polymer is water-soluble and conjugated via the linker to the ring moiety or the polymer is water-insoluble, wherein upon subcutaneous administration of said controlled-release CNP agonist a reduction or elimination of one or more side-effects is obtained compared to the subcutaneous administration of free D-H in an equivalent dosage, wherein the linker is a moiety -L1- of formula (II): wherein the dashed line indicates attachment by forming an amide bond with an amine side chain of a lysine of the CNP moiety; —X— is —C(R4R4a)—; —N(R4)13 ; —O—; —C(R4R4a)—C(R5R5a)—; —C(R5R5a)—C(R4R4a)—; —C(R4R4a)—N(R6)—; —N(R6)—C(R4R4a)—; —C(R4R4a)—O—; —O—C(R4R4a)—; or —C(R7R7a)—; X1 is C; or S(O); —X2— is —C(R8R8a)—; or —CR8R8a)—C(R9R9a)—; ═X3 is ═O; =S; or =N—CN; —R1, —R1a, —R2, —R2a, —R4, —R4a, —R5, —R5a, —R6, —R8, —R8a, —R9, —R9a are independently selected from the group consisting of —H; and C1-6 alkyl; —R3, —R3a are independently selected from the group consisting of —H; and C1-6 alkyl, provided that in case one of —R3, —R3aor both are other than —H they are connected to the N to which they are attached through a sp3-hybridized carbon atom; —R7 is —N(R10R10a); or NR10—(C═O)—R11 ; —R7a, —R10, —R10a, —R11 are independently of each other -H; or C1-6 alkyl; optionally, one or more of the pairs —R1a/—R4a, —R1a/—R5a, —R1a/—R7a, —R4a/—R5a, —R8a/—R9a form a chemical bond; optionally, one or more of the pairs —R1/—R1a, —R2/—R2a, —R3/—R3a, —R4/—R4a, —R5/—R5a, —R9/—R9a are joined together with the atom to which they are attached to form a C3-10 cycloalkyl; or 3- to 10-membered heterocyclyl; optionally, one or more of the pairs —R1/—R4, —R1/—R5, —R1/—R6, —R1/—R7a, —R4/—R5, —R4/—R6, —R8/—R9—, R2/—R3 are joined together with the atoms to which they are attached to form a ring A; optionally, —R3/—R3a are joined together with the nitrogen atom to which they are attached to form a 3- to 10-membered heterocycle; A is selected from the group consisting of phenyl; naphthyl; indenyl; indanyl; tetralinyl; C3-10 cycloalkyl; 3- to 10-membered heterocyclyl; and 8- to 11-membered heterobicyclyl; and wherein -Ll- is substituted with -L2-Z or -L2-Z′ and wherein -Ll- is optionally further substituted, provided that the hydrogen marked with the asterisk in formula (II) is not replaced by -L2-Z or -L2-Z′ or a substituent; wherein -L2- is a single chemical bond or a spacer; —Z is the water-soluble carrier; and —Z′ is the water-insoluble carrier. 2. The pharmaceutical composition of claim 1, wherein the side-effects are cardiovascular side-effects. 3. The pharmaceutical composition of claim 1, wherein the pharmaceutical composition comprises at least 25 nmol/ml of -D. 4. The pharmaceutical composition of claim 1, wherein the CNP moiety is CNP-38. 5. A method of treating a patient suffering from a disease that can be treated with a CNP agonist, the method comprising the step of subcutaneously administering to said patient the pharmaceutical composition of claim 1. 6. The method of claim 5, wherein the side-effect of hypotension is reduced compared to the subcutaneous administration of the free D-H in an equivalent dosage. 7. The pharmaceutical composition of claim 1, wherein the pharmaceutical composition comprises at least 100 nmol/ml of -D. 8. The method of claim 5, wherein the pharmaceutical composition is administered at a dose of at least 50 nmol/kg of -D. 9. The method of claim 5, wherein the pharmaceutical composition is administered at a dose of at least 100 nmol/kg of -D. 10. The method of claim 5, wherein administration of one unit dose of the pharmaceutical composition provides a pharmaceutically effective amount of D-H for at least 48 hours. 11. The method of claim 5, wherein administration of one unit dose of the pharmaceutical composition provides a pharmaceutically effective amount of D-H for at least 168 hours. 12. The method of claim 5, wherein the disease is selected from the group consisting of achondroplasia, hypochondroplasia, short stature, dwarfism, osteochondrodysplasias, thanatophoric dysplasia, osteogenesis imperfecta, achondrogenesis, chondrodysplasia punctata, homozygous achondroplasia, camptomelic dysplasia, congenital lethal hypophosphatasia, perinatal lethal type of osteogenesis imperfecta, short-rib polydactyly syndromes, rhizomelic type of chondrodysplasia punctata, Jansen-type metaphyseal dysplasia, spondyloepiphyseal dysplasia congenita, atelosteogenesis, diastrophic dysplasia, congenital short femur, Langer-type mesomelic dysplasia, Nievergelt-type mesomelic dysplasia, Robinow syndrome, Reinhardt syndrome, acrodysostosis, peripheral dysostosis, Kniest dysplasia, fibrochondrogenesis, Roberts syndrome, acromesomelic dysplasia, micromelia, Morquio syndrome, Kniest syndrome, metatrophic dysplasia, spondyloepimetaphyseal dysplasia, neurofibromatosis, Legius syndrome, LEOPARD syndrome, Noonan syndrome, hereditary gingival fibromatosis, neurofibromatosis type 1, cardiofaciocutaneous syndrome, Costello syndrome, SHOX deficiency, idiopathic short stature, growth hormone deficiency, osteoarthritis, cleidocranial dysostosis, craniosynostosis, dactyly, brachydactyly, camptodactyly, polydactyly, syndactyly, dyssegmental dysplasia, enchondromatosis, fibrous dysplasia, hereditary multiple exostoses, hypophosphatemic rickets, Jaffe-Lichtenstein syndrome, Marfan syndrome, McCune-Albright syndrome, osteopetrosis and osteopoikilosis. 13. The method of claim 5, wherein the disease is achondroplasia. 14. The method of claim 5, wherein the disease is selected from the group consisting of achondroplasia, hypochondroplasia, short stature, Noonan syndrome and SHOX deficiency. 15. The method of claim 5, wherein the pharmaceutical composition is administered at a dose of at least 2 nmol/kg of -D. 16. The method of claim 5, wherein administration of one unit dose of the pharmaceutical composition provides a pharmaceutically effective amount of D-H for at least 24 hours. |
