Application Number:US16/681,317
Patent Claims: 1. A method of using dextromethorphan with enhanced plasma levels to treat depression in a human being in need thereof, wherein the human being is an extensive metabolizer of the dextromethorphan and suffers from depression, comprising orally administering a dosage form comprising about 100 mg to about 200 mg of bupropion and about 40 mg to about 60 mg of the dextromethorphan to the human being, wherein the dosage form is administered once a day or twice a day for at least eight consecutive days, and the method results in a first Cmin of dextromethorphan that is at least about 20 ng/mL on the eighth day, and wherein the first Cmin of dextromethorphan is at least about 20 times a second Cmin of dextromethorphan that would result from administering the same amount of the dextromethorphan without the bupropion to the human being once a day or twice a day for eight consecutive days; wherein administering the dosage form is more effective in treating depression in the human being than administering the same amount of the bupropion alone.

2. The method of claim 1, wherein about 80 mg to about 100 mg of the dextromethorphan is orally administered daily.

3. The method of claim 1, wherein the weight ratio of the dextromethorphan to the bupropion in the dosage form is about 0.2 to about 0.4.

4. The method of claim 1, wherein the weight ratio of the dextromethorphan to the bupropion in the dosage form is about 0.3 to about 0.5.

5. The method of claim 1, wherein the weight ratio of the dextromethorphan to the bupropion in the dosage form is about 0.5 to about 0.7.

6. The method of claim 1, wherein the first Cmin of dextromethorphan is at least about 30 times the second Cmin of dextromethorphan.

7. The method of claim 1, wherein the first Cmin of dextromethorphan is at least about 40 times the second Cmin of dextromethorphan.

8. The method of claim 1, wherein the first Cmin of dextromethorphan is at least about 50 times the second Cmin of dextromethorphan.

9. The method of claim 1, wherein the first Cmin of dextromethorphan is at least about 30 ng/mL.

10. The method of claim 1, wherein the first Cmin of dextromethorphan is least about 50 ng/mL.

11. The method of claim 1, wherein about 200 mg of the bupropion is orally administered once daily or twice daily.

12. The method of claim 1, wherein about 200 mg to about 250 mg of the bupropion is orally administered daily.

13. The method of claim 1, wherein about 250 mg to about 300 mg of the bupropion is orally administered daily.

14. The method of claim 1, wherein the bupropion is deuterium-modified.

15. The method of claim 1, wherein the dextromethorphan is deuterium-modified.

16. The method of claim 1, wherein the dosage form is a solid.

17. The method of claim 1, wherein the dosage form further comprises a binder.

18. The method of claim 1, wherein the dosage form further comprises a disintegrating agent.

19. The method of claim 1, wherein the dosage form further comprises a lubricant.

20. The method of claim 1, wherein the dosage form is a tablet, a troche, a pill or a capsule.

21. The method of claim 1, wherein the dosage form is orally administered once a day or twice a day for at least 30 consecutive days.

22. The method of claim 1, wherein the dosage form is orally administered once a day or twice a day for at least 60 consecutive days.

23. The method of claim 1, wherein the human being is selected for suffering from treatment-resistant depression.

24. The method of claim 1, wherein the human being is selected for suffering from major depressive disorder.
