Summary for Patent: 7,510,873
Title:Postpartum cells isolated from umbilical cord tissue, and methods of making and using the same
Abstract: Cells derived from human umbilical cords are disclosed along with methods for their therapeutic use. Isolation techniques, culture methods and detailed characterization of the cells with respect to their cell surface markers, gene expression, and their secretion of trophic factors are described.

Assignee: Ethicon, Incorporated (Somerville, NJ)
Application Number:10/877,012
Patent Claims:1. An isolated cell, wherein said isolated cell is obtained by contacting mammalian umbilical cord tissue substantially free of blood with either dispase or dispase and at least one of hyaluronidase and collagenase and obtaining the cells yielded by said contacting step; wherein said isolated cell self-renews and expands its numbers in culture; wherein said isolated cell can differentiate into a cell that expresses markers indicative of differentiation into a type selected from the group consisting of neural cells, hepatocytes, adipocytes, pancreatic beta cells, chondrocytes, and cardiomyocytes; and wherein said isolated cell does not express CD45 or CD117.

2. A cell culture comprising the isolated cell of claim 1 wherein said culture expands in the presence of about 5% to about 20% oxygen.

3. The isolated cell of claim 1, wherein said isolated cell maintains a normal karyotype upon passaging.

4. The isolated cell of claim 1 wherein the isolated cell expresses one or more of CD10, CD13, CD44, CD73, CD90, PDGFr-alpha, or HLA-A,B,C.

5. The isolated cell of claim 4 wherein the isolated cell expresses each of CD10, CD13, CD44, CD73, CD90, PDGFr-alpha, and HLA-A,B,C.

6. The isolated cell of claim 1 wherein the isolated cell does not express one or more of CD31, CD34, CD141, or HLA-DR,DP,DQ, as detected by flow cytometry.

7. The isolated cell of claim 6 wherein the isolated cell expresses one or more of CD10, CD13, CD44, CD73, CD90, PDGFr-alpha, or HLA-A,B,C.

8. The isolated cell of claim 6 wherein the isolated cell does not express any of CD31, CD34, CD141, or HLA-DR,DP,DQ, as detected by flow cytometry.

9. The isolated cell of claim 8 wherein the isolated cell expresses each of CD10, CD13, CD44, CD73, CD90, PDGFr-alpha, and HLA-A,B,C.

10. A conditioned medium made by culturing the isolated cell of claim 1.

11. The conditioned medium of claim 10 comprising one or more of MCP- 1, IL-6, IL-8, GCP-2, HGF, KGF, FGF, HB-EGF, BDNF, TPO, and TIMP1.

12. A culture comprising the isolated cell of claim 1 and a mammalian cell that is not the isolated cell of claim 1.

13. The culture of claim 12 comprising a human cell line.

14. A three dimensional matrix comprising the isolated cell of claim 1.

15. The three-dimensional matrix of claim 14 wherein the matrix comprises a biocompatible or bioabsorbable polymer.

16. The three-dimensional matrix of claim 15 wherein the matrix comprises a nonwoven mat comprising a copolymer of glycolic and lactic acids, PCL/PGA copolymer, or self assembling peptides.

17. The isolated cell of claim 1 wherein said isolated cell requires L-valine for growth.

18. The isolated cell of claim 17 which doubles sufficiently to generate yields of greater than about 10.sup.14 cells in less than about 80 days in culture when seeded at about 10.sup.3 cells/cm.sup.2.

19. The isolated cell of claim 17 which doubles sufficiently to generate greater than about 10.sup.15 cells in less than about 80 days in culture when seeded at about 5 .times.10.sup.3 cells/cm.sup.2.

20. The isolated cell of claim 19 which doubles sufficiently to generate greater than about 10.sup.17 cells in less than about 65 days in culture when seeded at about 5 .times.10.sup.3 cells/cm.sup.2.

21. The isolated cell of claim 1 which undergoes at least 40 doublings in culture.

22. The isolated cell of claim 1, wherein said isolated cell is obtained from human umbilical cord tissue.

23. The isolated cell of claim 1 which attaches and doubles and expands when cultured on a coated or uncoated tissue culture vessel, wherein a coated tissue culture vessel comprises a coating with gelatin, laminin, collagen, polyornithine, polylysine, vitronectin, or fibronectin.

24. The cell culture of claim 2, which expands in a culture medium comprising from about 2% to about 15% serum, with or without beta-mercaptoethanol, and with or without a growth factor selected from the group consisting of EGF, FGF, PDGF, VEGF, IGF, and LIF.

25. A homogeneous cell population comprising isolated cells of claim 1.

26. The isolated cell of claim 1 wherein the collagenase is a collagenase from Clostridium histolyticum.
