Title: Islet isolation process
Abstract:A method of producing intact islets of Langerhans in an insulin producing condition uses a mixture of Hank\'s solution and 10% by volume fetal calf serum to ductally distend the human pancreas. The exocrine tissue of the pancreas is digested at about 37.degree. C. by an enzyme preparation of collagenase, trypsin and proteolytic enzyme present in the mixture at a level of about 0.2% by weight. The digestion is monitored at regular intervals during the process. The digested pancreas is comminuted, filtered and intact islets of Langerhans are recovered. The recovered islets retain their insulin producing properties.

Patent Claims:1. The method of recovering an in vitro islet collection containing isolated islets of Langerhans from a pancreas, said method comprising distending the pancreas with an enzyme preparation comprising collagenase, disrupting at least a portion of the previously distended pancreas by mechanical disruption to break down the exocrine tissue of the pancreas and free islets, substantially isolating the freed islets from the exocrine tissue of the pancreas, and recovering at least about 66,600 isolated morphologically and functionally intact viable islets.

2. The method of claim 1 in which said islets secrete insulin at a basal rate of at least about 0.9.mu. units/islet/minute and at a stimulated rate of at least about 2.2.mu. units/islet/minute.

3. The method of claim 1 in which said pancreas is derived from pig pancreas.

4. The method of claim 1 in which said pancreas is derived from human pancreas.

5. The method of claim 1 in which said pancreas is derived from a large animal pancreas.

6. The method of claim 1 in which the islets in said recovered islet collection contain no more than about 5% pancreatic tissue impurities.
