| Abstract: | The disclosure provides, in part, follistatin polypeptides that are suitable for use in local administration and methods for use. |
| Inventor(s): | Kumar; Ravindra (Acton, MA), Grinberg; Asya (Lexington, MA) |
| Assignee: | ACCELERON PHARMA INC. (Cambridge, MA) |
| Application Number: | 14/731,009 |
| Patent Claims: | 1. A polypeptide comprising an amino acid sequence selected from: a) the amino acid sequence of SEQ ID NO: 43, and b) the amino acid sequence of SEQ ID NO: 43, wherein the final
(carboxy-terminal) lysine (K) of SEQ ID NO: 43 is absent.
2. The polypeptide of claim 1, wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 43. 3. The polypeptide of claim 2, wherein the polypeptide is in a homodimer. 4. The polypeptide of claim 1, wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 43, wherein the final (carboxy-terminal) lysine (K) of SEQ ID NO: 43 is absent. 5. The polypeptide of claim 4, wherein the polypeptide is in a homodimer. 6. A polypeptide comprising an amino acid sequence selected from: a) the amino acid sequence of SEQ ID NO: 42, and b) the amino acid sequence of SEQ ID NO: 42, wherein the final (carboxy-terminal) lysine (K) of SEQ ID NO: 42 is absent. 7. The polypeptide of claim 6, wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 42. 8. The polypeptide of claim 7, wherein the polypeptide is in a homodimer. 9. The polypeptide of claim 6, wherein the polypeptide comprises the amino acid sequence of SEQ ID NO: 42, wherein the final (carboxy-terminal) lysine (K) of SEQ ID NO: 42 is absent. 10. The polypeptide of claim 9, wherein the polypeptide is in a homodimer. 11. A polypeptide comprising a first amino acid sequence and a second amino acid sequence, wherein the first amino acid sequence consists of an amino acid sequence selected from the group consisting of SEQ ID NO: 15 and 16, wherein the second amino acid sequence comprises a constant domain of an immunoglobulin, wherein a linker directly connects the C-terminal portion of the first amino acid sequence to the N-terminal portion of the second amino acid sequence, and wherein the linker is 1-10 amino acids in length. 12. The polypeptide of claim 11, wherein the linker comprises the sequence of SEQ ID NO: 46. 13. A fusion protein comprising a first amino acid sequence derived from follistatin that consists of the amino acid sequence of SEQ ID NO: 15 or 16; and a second amino acid sequence that is heterologous to follistatin and comprises a constant domain of an immunoglobulin G (IgG), wherein the constant domain of the IgG is selected so as to mediate no substantial antibody-dependent cell-mediated cytotoxicity (ADCC) or no substantial complement dependent cytotoxicity (CDC), wherein a linker directly connects the C-terminal portion of the first amino acid sequence to the N-terminal portion of the second amino acid sequence, and wherein the linker is 1-10 amino acids in length. 14. The fusion protein of claim 13, wherein the linker comprises the sequence of SEQ ID NO: 46. 15. The fusion protein of claim 13, wherein the constant domain of the IgG is a constant domain of an IgG1. 16. The fusion protein of claim 15, wherein the constant domain of the IgG1 comprises the amino acid sequence of SEQ ID NO: 17. 17. The fusion protein of claim 13, wherein the constant domain of the IgG is a constant domain of an IgG2. 18. The fusion protein of claim 17, wherein the constant domain of the IgG2 comprises the amino acid sequence of SEQ ID NO: 18. 19. The fusion protein of claim 18, wherein the linker comprises the sequence of SEQ ID NO: 46. |
