Title:Stabilized compositions of volatile alkylating agents and methods of using thereof
Abstract: A composition and method for treatment of cancer. The composition for treating a skin disorder, comprising: a Nitrogen Mustard or an HX salt of the Nitrogen Mustard, wherein the Nitrogen Mustard or the HX salt of the Nitrogen Mustard is in a non-aqueous vehicle or carrier that does not include petrolatum or ethanol, wherein the non-aqueous vehicle or carrier that does not include petrolatum or ethanol does not include petrolatum or ethanol. The method comprises topically applying the composition of a Nitrogen Mustard or a HX salt of the Nitrogen Mustard to the affected skin, wherein the Nitrogen Mustard or the HX salt of the Nitrogen Mustard is in a non-aqueous vehicle or carrier that does not include petrolatum or ethanol, wherein the non-aqueous vehicle or carrier does not include petrolatum or ethanol.

Claims:1. A topical composition comprising: (a) a bis-(2-chloroethyl)methylamine or a pharmaceutically acceptable salt thereof, wherein the bis-(2-chloroethyl)methylamine or pharmaceutically acceptable salt thereof is present in an amount of at least about 0.001% by weight of the composition; and (b) a non-aqueous vehicle or carrier that does not include petrolatum or ethanol, wherein the non-aqueous vehicle or carrier comprises (i) a compound of formula HOCH.sub.2CH.sub.2OCH.sub.2CH.sub.2OR.sub.79, wherein R.sub.79 is a linear alkyl group having from 1 to 6 carbon atoms; (ii) hydroxypropylcellulose; (iii) butylated hydroxytoluene, benzyl alcohol, sodium benzoate, parabens, or edetate disodium; and (iv) a secondary or tertiary alcohol having from 1 to 20 carbon atoms.

2. The topical composition of claim 1, wherein the bis-(2-chloroethyl)methylamine or pharmaceutically acceptable salt thereof is in the form of an acid-addition or base-addition salt.

3. The topical composition of claim 2, wherein the bis-(2-chloroethyl)methylamine or pharmaceutically acceptable salt thereof is in the form of an acid-addition salt.

4. The topical composition of claim 3, wherein the acid-addition salt is an HCl, HBr, HI, H.sub.2SO.sub.4, or HNO.sub.3 salt.

5. The topical composition of claim 4, wherein the acid-addition salt is an HCl salt.

6. The topical composition of claim 1, wherein the secondary or tertiary alcohol is the secondary alcohol.

7. The topical composition of claim 6, wherein the secondary alcohol is isopropyl alcohol.

8. The topical composition of claim 1, wherein the secondary or tertiary alcohol is present in an amount of from 1 to 20 percent by weight of the composition.

9. The topical composition of claim 1, wherein in the compound of formula HOCH.sub.2CH.sub.2OCH.sub.2CH.sub.2OR.sub.79, R.sub.79 is a linear alkyl group having 2 carbon atoms.

10. The topical composition of claim 9, wherein the compound of formula HOCH.sub.2CH.sub.2OCH.sub.2CH.sub.2OR.sub.79 is present in an amount of from 15 to 60 percent by weight of the composition.

11. The topical composition of claim 1, wherein the hydroxypropylcellulose is present in an amount of at most 5 percent by weight of the composition.

12. The topical composition of claim 1, wherein the butylated hydroxytoluene is present in an amount of at most 1 percent by weight of the composition.

13. The topical composition of claim 1, wherein the edetate disodium is present in an amount of at most 0.05 percent by weight of the composition.

14. The topical composition of claim 1, wherein the non-aqueous vehicle or carrier further comprises a menthol in an amount of at most 1 percent by weight of the composition.

15. The topical composition of claim 1, wherein the non-aqueous vehicle or carrier further comprises a propylene glycol in an amount of from 15 to 60 percent by weight of the composition.

16. The topical composition of claim 1, wherein the non-aqueous vehicle or carrier further comprises an organic acid in an amount of from 1 to 15 percent by weight of the composition, wherein the organic acid is selected from the group consisting of oxalic acid, p-bromophenylsulfonic acid, carbonic acid, succinic acid, citric acid, benzoic acid, 2-acetoxybenzoic acid, acetic acid, phenylacetic acid, propionic acid, glycolic acid, stearic acid, lactic acid, malic acid, tartaric acid, ascorbic acid, maleic acid, hydroxymaleic acid, glutamic acid, salicylic acid, sulfanilic acid, fumaric acid, and combinations thereof.

17. The topical composition of claim 16, wherein the organic acid is lactic acid.

18. The topical composition of claim 1, comprising: (a) the bis-(2-chloroethyl)methylamine or pharmaceutically acceptable salt thereof is an HCl salt and is present in an amount of at least about 0.001% by weight of the composition; and (b) the non-aqueous vehicle or carrier comprises: (i) the HOCH.sub.2CH.sub.2OCH.sub.2CH.sub.2OR.sub.79 in an amount of from 15 to 60 percent by weight of the composition, wherein R.sub.79 is a linear alkyl group having 2 carbon atoms; (ii) the hydroxypropylcellulose in an amount of at most 5 percent by weight of the composition; (iii) the butylated hydroxytoluene in an amount of at most 1 percent by weight of the composition and the edetate disodium in an amount of at most 0.05 percent by weight of the composition; and (iv) the secondary or tertiary alcohol in an amount of from 1 to 20 percent by weight of the composition, wherein the secondary or tertiary alcohol is isopropyl alcohol.
