Title: | Polymorphic crystalline forms of tiacumicin B |
Abstract: | The invention relates to novel forms of compounds displaying broad spectrum antibiotic activity, especially crystalline polymorphic forms and amorphous forms of such compounds, compositions comprising such crystalline polymorphic forms and amorphous forms of such compounds, processes for manufacture and use thereof. The compounds and compositions of the invention are useful in the pharmaceutical industry, for example, in the treatment or prevention of diseases or disorders associated with the use of antibiotics, chemotherapies, or antiviral therapies, including, but not limited to, colitis, for example, pseudo-membranous colitis; antibiotic associated diarrhea; and infections due to Clostridium difficile ("C. difficile"), Clostridium perfringens ("C. perfringens"), Staphylococcus species, for example, methicillin-resistant Staphylococcus, or Enterococcus including Vancomycin-resistant enterococci. |

Claims: | 1. A polymorphic form of a compound of Formula I: ##STR00010## characterized by a powder x-ray diffraction pattern wherein said x-ray diffraction pattern comprises peaks at diffraction angles 2.theta. of 7.7.degree., 15.0.degree., and 18.8.degree..+-.0.2 as said peaks are set forth in FIG. 1. 2. A solid dosage form comprising the polymorphic form of a compound of Formula I of claim 1. 3. The solid dosage form of claim 2, wherein the polymorphic form of a compound of Formula I is present in at least about 75% to about 99.99% of the total weight. 4. The solid dosage form of claim 2, wherein the polymorphic form of a compound of Formula I is present in at least about 85% of the total weight. 5. The solid dosage form of claim 2, wherein the polymorphic form of a compound of Formula I is present in at least about 90% of the total weight. 6. The solid dosage form of claim 2, wherein the polymorphic form of a compound of Formula I is present in at least about 95% of the total weight. 7. The solid dosage form of claim 2, wherein the polymorphic form of a compound of Formula I is present in at least about 99% of the total weight. 8. The polymorphic form of the compound of Formula I according to claim 1 characterized by a DSC endotherm in the range of about 174.degree. C. to about 186.degree. C. 9. A solid dosage form comprising the polymorphic form of a compound of Formula I of claim 8. 10. The solid dosage form of claim 9, wherein the polymorphic form of a compound of Formula I is present from about 75% to about 99.99% of the total weight. 11. A polymorphic form of a compound of Formula I: ##STR00011## characterized by: (i) a powder x-ray diffraction pattern wherein said x-ray diffraction Pattern comprises peaks at diffraction angles 2.theta. of 7.7.degree., 15.0.degree., and 18.8.degree..+-.0.2 as said peaks are set forth in FIG. 1; and (ii) a DSC endotherm in the range of about 174.degree. C. to about 186.degree. C. 12. A solid dosage form comprising the polymorphic form of a compound of Formula I of claim 11. 13. The solid dosage form of claim 12 wherein the polymorphic form of a compound of Formula I is present from about 75% to about 99.99% of the total weight. 14. The solid dosage form of claim 12, wherein the polymorphic form of a compound of Formula I is present in about 90% of the total weight. 15. A pharmaceutical composition comprising the solid dosage form of claim 2. 16. A pharmaceutical composition comprising the solid dosage form of claim 9. 17. The pharmaceutical composition of claim 15 further comprising a pharmaceutically acceptable excipient. 18. The pharmaceutical composition of claim 16 further comprising a pharmaceutically acceptable excipient. 19. The polymorphic form of the compound of Formula I of claim 1 characterized by a diffraction pattern as set forth in FIG. 1. 20. The polymorphic form of the compound of Formula I of claim 11 characterized by a diffraction pattern as set forth in FIG. 1. |
