Summary for Patent: 9,155,763
Title:Lgr5+ somatic stem cells
Abstract: Described is an isolated somatic stem cell that is 2 to less than 6 micrometer in size and Lgr5+. Methods of preparing and using the cell are also described.
Inventor(s): Wang; James (Monterey Park, CA)
Assignee: StemBios Technologies, Inc. (Monterey Park, CA)
Application Number:14/097,572
Patent Claims:1. An isolated somatic stem cell that is 2 to less than 6 micrometer in size and Lgr5+.

2. The isolated somatic stem cell of claim 1, wherein the isolated somatic stem cell is pluripotent or totipotent.

3. The isolated somatic stem cell of claim 1, wherein the cell is a human cell.

4. The isolated somatic stem cell of claim 1, wherein the cell is non-adherent.

5. The isolated somatic stem cell of claim 1, wherein the cell is CD9- CD349-.

6. The isolated somatic stem cell of claim 5, wherein the cell is Oct4+, Nanog+, CD133-, CD66e-, and Sox2-.

7. A method of preparing a population of somatic stem cells, the method comprising, obtaining from a subject a tissue sample containing a plurality of cells, incubating the sample with EDTA or heparin in a container until the sample is separated into an upper layer and a lower layer, collecting the upper layer, and isolating from the upper layer a population of somatic stem cells that are 2 to less than 6 micrometers in size and Lgr5+.

8. The method of claim 7, wherein the tissue sample is blood, bone marrow, skeletal muscle, or adipose tissue.

9. The method of claim 7, further comprising, between the obtaining step and the incubating step, digesting the tissue sample with a collagenase, wherein the tissue sample is skeletal muscle or adipose tissue.

10. The method of claim 7, wherein the subject is a human.

11. The method of claim 7, wherein the isolating step is conducted by flow cytometry.

12. The method of claim 7, further comprising, after the collecting step, incubating the upper layer with ADP.

13. The method of claim 7, wherein the incubating step is performed by incubating the sample with heparin, and, after the collecting step and before the isolating step, the collected upper layer is incubated with EDTA.

14. A population of somatic stem cells prepared by the method of claim 7.

15. The population of claim 14, wherein the somatic stem cells are CD9- CD349-.

16. The population of claim 15, wherein the somatic cells are Oct4+, Nanog+, CD133-, CD66e-, and Sox2-.

17. The population of claim 14, wherein the population is prepared from a blood or a bone marrow sample obtained from a subject.

18. The population of claim 14, wherein the subject is a human.

19. The population of claim 14, wherein the somatic stem cells are non-adherent.

20. A cell bank comprising a plurality of populations of somatic stem cells, the plurality of populations being prepared by the method of claim 7 from blood or bone marrow samples obtained from different subjects.

21. A method of generating hepatocytes, neuronal cells, or skeletal muscle cells in a subject, the method comprising administering to the subject a population of somatic stem cells prepared by the method of claim 7, whereby the somatic stem cells differentiate into hepatocytes, neuronal cells, or skeletal muscle cells in the subject.

22. A method of generating hepatocytes, neuronal cells, or adipocytes, the method comprising culturing a population of somatic stem cells prepared by the method of claim 7 under conditions suitable for hepatocytic, neuronal, or adipocytic differentiation, whereby hepatocytes, neuronal cells, or adipocytes are generated, respectively.
