Abstract: One embodiment provides a method for enzymatic treatment, including the steps of forming a closed space on a local tissue area with a device and infusing an enzyme solution into the closed space for enzymatic treatment. The method according to the embodiment is capable of treating the local tissue area with enzymes for enhancing cell proliferation in the treated tissue area and preventing damage of the adjacent normal tissues. A device and kit used for the method are also provided.

Patent Claims:1. A device for enzymatic treatment, comprising: a hollow cavity, comprising a first side and a second side opposed to the first side, wherein the first and second sides comprise at least one opening, wherein the device forms a closed space on a tissue surface, and wherein the second side comprises two flanks, and each flank comprises one extrusion thereon.

2. The device as claimed in claim 1, further comprising: an infusion element, wherein the infusion element comprises a container part and an infusion opening configured to infuse an enzyme solution through the first side and into the closed space.

3. The device as claimed in claim 1, further comprising: an extraction element, wherein the extraction element comprises an extraction part and an extraction opening for extracting fluids through the second and first sides and out from the closed space.

4. The device as claimed in claim 1, wherein each extrusion extends 3-5 mm from the respective flank of the second side.

5. The device as claimed in claim 1, wherein each extrusion is 3-5 mm from the end of the second side.

6. The device as claimed in claim 1, further comprising: an auxiliary element, wherein the auxiliary element comprises a cap body and an opening for assisting the immobilization of the device on the tissue surface.

7. The device as claimed in claim 1, wherein the second side comprises a biocompatible material.

8. The device as claimed in claim 7, wherein the biocompatible material comprises polyether ether ketone (PEEK), stainless steel or silicone.

9. The device as claimed in claim 1, wherein the tissue comprises cartilage tissues, bone tissues or nerve tissues.

10. The device as claimed in claim 1, further comprising an enzyme solution comprising one or more enzymes selected from the group consisting of a collagenase, a hyaluronidase, a trypsin, a lipase and a proteinase.

11. A kit for treatment of tissue defects, comprising the device as claimed in claim 1 and an enzyme solution.

12. The kit as claimed in claim 11, wherein the enzyme solution comprises one or more enzymes selected from the group consisting of a collagenase, a hyaluronidase, a trypsin, a lipase and a proteinase.

13. The kit as claimed in claim 11, wherein the concentration of the enzyme solution is 2-5 U/ml.
