Abstract: The present invention is in the field of plant genetics and provides recombinant nucleic acid molecules, constructs, and other agents associated with the coordinate manipulation of multiple genes in the fatty acid synthesis pathway. In particular, the agents of the present invention are associated with the simultaneous enhanced expression of certain genes in the fatty acid synthesis pathway and suppressed expression of certain other genes in the same pathway. Also provided are plants incorporating such agents, and in particular plants incorporating such constructs where the plants exhibit altered seed oil compositions.

Patent Claims:1. A recombinant nucleic acid molecule comprising a recombinant FAD2 sequence and a recombinant FATB sequence, wherein said recombinant FAD2 sequence consists of less than the entire sequence of a soybean FAD2-1 intron that is at least 95% identical to SEQ ID NO: 1 and is between 20 to about 420 contiguous nucleotides in length and said recombinant FATB sequence is a soybean FATB-1 sequence between 40 and 500 nucleotides in length, and said soybean FATB-1 sequence is at least 95% identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37, or said soybean FATB-1 sequence is a combination of sequences that is selected from the group consisting of a 25 contiguous nucleotide sequence that is at least 95% identical to SEQ ID NOs: 28 to 37, wherein said recombinant nucleic acid molecule when expressed in a soybean plant cell suppresses the expression of endogenous soybean FAD2-1 and FATB-1.

2. The recombinant nucleic acid molecule of claim 1, wherein said less than the entire sequence of a soybean FAD2-1 intron is a nucleic acid sequence that is at least 97% identical to a fragment of a soybean FAD2-1A intron.

3. The recombinant nucleic acid molecule of claim 1, wherein said less than the entire sequence of a soybean FAD2-1 intron is identical to SEQ ID NO: 1 and is between 20 to about 420 contiguous nucleotides in length.

4. The recombinant nucleic acid molecule of claim 1, wherein said less than the entire sequence of a soybean FAD2-1 intron is truncated or deleted at the 5' end, at the 3' end, or internally.

The recombinant nucleic acid molecule of claim 1, wherein said fragment of a soybean FATB gene is a combination of two FATB-1 nucleic acid sequences at least 95% identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37 or said soybean FATB-1 sequence is a combination of sequences that is selected from the group consisting of a 25 contiguous nucleotide sequence that is at least 95% identical to SEQ ID NOs: 28 to 37.

6. The recombinant nucleic acid molecule of claim 5, wherein said combination of two FATB-1 nucleic acid sequences is between about 40 to about 450nucleotides in length.

The recombinant nucleic acid molecule of claim 5, wherein said combination of two FATB-1 nucleic acid sequences is selected from the group consisting of a 25contiguous nucleotide sequence that is at least 95% identical to soybean FATB-1 3' UTR of SEQ ID NO:36, a soybean FATB-1 5' UTR of SEQ ID NO:37, and a soybean FATB-1 of SEQ ID NO:28 from position 1122 to 1290.

8. The recombinant nucleic acid molecule of claim 7, wherein said soybean FATB-1 3' UTR is identical to SEQ ID NO:36.

9. The recombinant nucleic acid molecule of claim 7, wherein said soybean FATB-1 5' UTR is identical to SEQ ID NO:37.

The recombinant nucleic acid molecule of claim 1, further comprising one or more promoters functional in a plant cell selected from the group consisting of a 7S.alpha. promoter, a 7S.alpha. a' promoter, a CaMV 35S promoter, an FMV promoter, a napin promoter, a FAD2-1A promoter, and a bean arcelin promoter.

11. The recombinant nucleic acid molecule of claim 10, wherein said one or more promoters is a 7S.alpha. ' promoter.

12. The recombinant nucleic acid molecule of claim 1, further comprising a genetic element selected from the group consisting of a 5' regulatory region, a 3'transcriptional terminator, a 3' polyadenylation signal, a transit sequence, a targeting sequence, an enhancer, an operator, and a screenable marker.

The recombinant nucleic acid molecule of claim 1, wherein said soybean FAD2-1 intron that is at least 95% identical to SEQ ID NO: 1 and is between about 20and about 420 contiguous nucleotides in length is selected from the group consisting of between about 30 to about 420, between about 40 to about 320, between about 50 to about 200, between about 50 to about 400, between about 50 to about 420, between about 60 to about 320, between about 70 to about 220, between about 100 to about 200, between about 150 to about 200, between about 150 to about 220, between about 150 to about 400, between about 200 to about 300, and between about 300 to about 400 contiguous nucleotides in length.

14. The recombinant nucleic acid molecule of claim 1, wherein said soybean FAD2-1 intron is 320 nucleotides in length.

The recombinant nucleic acid molecule of claim 1, wherein said combination of soybean FATB-1 sequences is about 80 to about 450 contiguous nucleotides, about 100 to about 500 contiguous nucleotides, about 70 to about 500 contiguous nucleotides, about 200 to about 400 contiguous nucleotides, about 150 to about 300 contiguous nucleotide, about 250 to about 350 contiguous nucleotides, or about 200 to about 350 contiguous nucleotides of said any FATB-1 sequence.

16. The recombinant nucleic acid molecule of claim 15, wherein at least one sequence of said soybean FATB-1 sequence combination is selected from the group consisting of about 25 contiguous nucleotides of SEQ ID NO:28, about 25 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, about 30 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, about 35 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 38 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 39 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 40contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 41 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 42 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, 43 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, about 45 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, about 50 contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, about 55contiguous nucleotides in length of a FATB 5' UTR of SEQ ID NO:37, and about 60 contiguous nucleotides of a FATB 5' UTR of SEQ ID NO:37.

17. The recombinant nucleic acid molecule of claim 1, wherein said recombinant molecule is pMON95829.

18. A transformed soybean plant, or a part thereof, comprising the recombinant nucleic acid molecule of claim 1.

19. The transformed soybean plant, or a part thereof, according to claim 18, wherein a seed of said plant has an oleic acid content of about 42% to about 85% by weight of the total fatty acids and a saturated fatty acid content of less than about 8% by weight of the total fatty acids.

20. The transformed soybean plant, or a part thereof, of claim 19, wherein said oleic acid content is selected from the group consisting of about 50% to about 80%, about 55% to about 80%, about 55% to about 75%, about 55% to about 65%, about 60% to about 80%, about 65% to about 80%, about 60% to about 85%, and about 46% to about 75% by weight of the total fatty acids.

21. The transformed soybean plant, or a part thereof, of claim 19, wherein said saturated fatty acid content of less than about 8% by weight of the total fatty acids is selected from the group consisting of about 7% or less, about 6% or less, about 5% or less, about 4.5% or less, about 4% or less, about 3.5% or less, about 3% or less, about 2.5% or less, and about 2% or less.

22. The transformed soybean plant, or a part thereof, of claim 19, wherein said saturated fatty acid content of less than about 8% by weight of the total fatty acids is a range selected from the group consisting of from 0.5 to 2%, 0.5 to 3%, 0.5 to 4.5%, 0.5 to 6%, 0.5 to 7%, 0.5 to 8%, 1 to 4%, 1 to 5%, 1 to 6%, 1 to 7%, 1.5 to 5%, 1.5 to 6%, 1.5 to 7%, 2 to 5%, 2 to 6%, 2 to 7%, 3 to 5%, 3 to 6%, 3 to 7%, 4 to 7%, and 5 to 7%.

23. The transformed soybean plant, or a part thereof, of claim 18, wherein said plant part is selected from the group consisting of a feed preparation, meal preparation, protein preparation, oil preparation, seed, endosperm, ovule, pollen, root, tuber, stem, leaf, stalk, pod, and flower.

24. The transformed soybean plant, or a part thereof, of claim 23, wherein said plant part is an oil preparation.

25. The transformed soybean plant, or a part thereof, of claim 24, wherein said oil preparation comprises an oleic acid content selected from the group consisting of about 50% to about 80%, about 55% to about 80%, about 55% to about 75%, about 55% to about 65%, about 60% to about 80%, about 65% to about 80%, about 60% to about 85%, and about 46% to about 75% by weight of the total fatty acids.

26. The transformed soybean plant, or a part thereof, of claim 24, wherein said oil preparation comprising a saturated fatty acid content of less than about 8% by weight of the total fatty acids is selected from the group consisting of about 7% or less, about 6% or less, about 5% or less, about 4.5% or less, about 4% or less, about 3.5% or less, about 3% or less, about 2.5% or less, and about 2% or less.

27. The transformed soybean plant, or a part thereof, of claim 23, wherein said plant part is a meal preparation.

28. The recombinant nucleic acid molecule of claim 1, wherein said soybean FATB-1 sequence that is at least 95% identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37 is truncated or deleted at the 5' end, at the 3' end, or internally.

29. The recombinant nucleic acid molecule of claim 1, wherein said soybean FATB-1 sequence is at least 97% identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37, or said soybean FATB-1 sequence is a combination of soybean FATB-1 sequences is selected from the group consisting of a 25contiguous nucleotide sequence that is at least 97% identical to SEQ ID NOs: 28 to 37.

30. The recombinant nucleic acid molecule of claim 29, wherein said soybean FATB-1 sequence is at least 99% identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37 or said soybean FATB-1sequence is a combination of soybean FATB-1 sequences selected from the group consisting of a 25 contiguous nucleotide sequence that is at least 99% identical to SEQ ID NOs: 28 to 37.

31. The recombinant nucleic acid molecule of claim 30, wherein said soybean FATB-1 sequence is identical to 25 contiguous nucleotides of a sequence selected from the group consisting of SEQ ID NOs: 28 to 37 or said soybean FATB-1 sequence is a combination of soybean FATB-1 sequences selected from the group consisting of a 25contiguous nucleotide sequence that is identical to SEQ ID NOs: 28 to 37.

32. The recombinant nucleic acid molecule of claim 30, wherein said soybean FATB-1 sequence is a fragment of a soybean FATB-1 gene comprising a 192nucleotide sequence at least 95% identical to SEQ ID NO: 28 from position 274 to position 296and position 1122 to 1290.
