Abstract: The invention relates to new asparaginases having improved properties, preferably improved thermotolerance, such as improved activity at high temperatures and/or improved thermostability. The invention also relates to DNA sequences encoding such improved asparaginases, their production in a recombinant host cell, as well as methods of using the asparaginases, in particular for reduction of acrylamide in foods. The invention furthermore relates to methods of generating and preparing asparaginase variants having improved properties.

Patent Claims:1. A polypeptide which: (a) has asparaginase activity; (b) has at least 90% identity to amino acids 80 to 378 of SEQ ID NO: 1; and (c) comprises an amino acid difference compared to SEQ ID NO: 1 at a position corresponding to any of positions 83-84, 196, 226, 278, 290, 312, 336-337 and/or 366 in SEQ ID NO: 1.

2. The polypeptide of claim 1, wherein the amino acid difference is a substitution.

3. The polypeptide of claim 2, which comprises a substitution in at least one of the following positions: 196, 278, 290, 312, 336 and/or 366.

4. The polypeptide of claim 2, which comprises at least one of the following substitutions: 83PN, 84P/D, 196E/I, 278H/K/P/Q/R, 290E/L/V, 312N/R/V/Y, 336C/G/L/P, 337F/I/K/Q/R, and/or 366P.

5. The polypeptide of claim 2, which comprises at least one of the following substitutions: 83V, 84D, 196I, 278H/Q, 290V, 312Y, 336C/G/L, 337F/I and/or 366P.

6. The polypeptide of claim 1, which further comprises a substitution in at least one of the following positions: 54, 57, 68-74, 82, 85-86, 88, 93-96, 102, 107, 111, 113, 115, 137, 139, 164, 165, 172, 176, 184-186, 194, 201, 206, 209, 212, 214, 215, 219, 220, 223, 224, 228, 231, 235, 246, 249, 255, 260, 262, 264, 266, 271, 275, 279-281, 283, 286, 288, 299, 306, 307, 309-321, 323, 325, 327-335, 338-342, 349, 351, 353, 356-363, 365 and/or 375.

7. The polypeptide of claim 1, which further comprises a substitution at position 70.

8. The polypeptide of claim 6, which further comprises a substitution in at least one of the following positions: 54, 57, 86, 102, 137, 164, 201, 228, 260, 262, 283, 307, 323, 327, 334, 349, 351, 353 and/or 375.

11. The polypeptide of claim 6, which comprises one of the following substitutions: N70H, N70K or N70S.

12. The polypeptide of claim 6, which comprises the substitution N70K.

13. The polypeptide of claim 6, which comprises one of the following substitutions: 323R, 327V, 349Q, 351A and/or 353I.

14. The polypeptide of claim 1, which has at least 95% identity to amino acids 80 to 378 of SEQ ID NO: 1.

15. The polypeptide of claim 1, which has at least 98% identity to amino acids 80 to 378 of SEQ ID NO: 1.

16. An isolated nucleic acid sequence comprising a nucleic acid sequence which encodes the polypeptide of claim 1.

17. A nucleic acid construct comprising a nucleic acid sequence of claim 16 operably linked to one or more control sequences that direct the production of the polypeptide in a suitable expression host.

18. A recombinant expression vector comprising the nucleic acid construct of claim 17.

19. A recombinant host cell comprising the expression vector of claim 18.

20. A method for producing a polypeptide, the method comprising: (a) cultivating the host cell of claim 19 to produce a supernatant comprising the polypeptide; and (b) recovering the polypeptide.

21. A method for producing a food product, comprising: (a) providing a food material; and (b) treating the food material with the polypeptide of claim 1.

22. The method of claim 21, wherein the food material is heated after step (b).

23. The method of claim 21, wherein the method is for reduction of acrylamide in the food product.

24. The method of claim 21, wherein the food product is a cereal based product; a vegetable based product; or a coffee based product.

25. The method of claim 21, wherein the food product is bread, pastry, cake, pretzels, bagels, cookies, gingerbread, gingercake, breakfast cereals, crispbread, or a potato based product.

26. The method of claim 25, wherein the potato based product is french fries, potato chips or crisps, fabricated potato snacks or croquettes.
