Abstract:A novel protein allergen Der p VII of Dermatophagoides pteronyssinus is described. A cDNA clone encoding Der p VII was isolated from a .lambda.gt11 library of D. pteronyssinus cDNA. The nucleic acid sequence of Der p VII encodes a 198 residue mature processed protein having a predicted molecular weight of 22,177 daltons. Der p VII protein may be used as the active ingredient in therapeutic composition for the treatment of sensitivity to house dust mites. The protein may also be used in methods of diagnosing such sensitivity.

Patent Claims:1. An isolated nucleic acid sequence coding for a house dust mite allergen Der p VII comprising bases 119-715 of the nucleotide sequence of FIG. 3, SEQ ID NO: 1.

2. An isolated nucleic acid sequence for a house dust mite allergen Der p VII, said nucleic acid sequence comprising the nucleotide sequence of FIG. 3, SEQ ID NO: 1, or the coding region thereof.

3. An expression vector coding for a house dust mite allergen Der p VII comprising a nucleic acid sequence of claim 2 operably linked to a regulatory sequence.

4. An expression vector coding for a house dust mite allergen Der p VII comprising a nucleic acid sequence of claim 1 operably linked to a regulatory sequence.

5. A host cell containing an expression vector of claim 3.

6. A host cell of claim 5 wherein the cell is eukaryotic.

7. A method of producing Der p VII protein, comprising: culturing a host cell containing a vector directing expression of a nucleic acid sequence coding for Der p VII protein, said nucleic acid sequence comprising the nucleotide sequence of FIG. 3, SEQ ID NO: 1, or the coding region thereof, under conditions appropriate for expression of Der p VII protein; and isolating Der p VII protein from the culture.

8. A host cell containing an expression vector of claim 4.

A method of producing Der p VII protein, comprising: culturing a host cell containing a vector directing expression of a nucleic acid sequence coding for Der p VII protein, said nucleic acid sequence comprising bases 119-715 of the nucleotide sequence of FIG. 3, SEQ ID NO: 1, under conditions appropriate for expression of Der p VII protein; and isolating Der p VII protein from the culture.

10. An isolated nucleic acid sequence encoding a house dust mite allergen, Der p VII, said allergen comprising the amino acid sequence shown in FIG. 3, SEQ ID NO: 2.

11. An isolated nucleic acid sequence encoding a house dust mite allergen, Der p VII, said allergen comprising residues 18-215 of the amino acid sequence shown in FIG. 3, SEQ ID NO: 2.

12. An isolated nucleic acid sequence coding for a protein allergen capable of stimulating T cells specific for a Group VII allergen of Dermatophagoides pteronyssinus, Der p VII, wherein the Der p VII allergen comprises the amino acid sequence shown in FIG. 3 (SEQ ID NO: 2).
