Summary for Patent: 6,333,189
Title: Method of making an electrotransport device
Abstract:Method of making an electrotransport device containing an analog of a parent polypeptide having one or more amino acid residues substituted relative to the parent polypeptide with an amino acid residue selected from the group consisting of proline, glycine and asparagine.
Inventor(s): Holladay; Leslie A. (Mountain View, CA), Oldenburg; Kevin R. (Chaddsford, PA)
Assignee: ALZA Corporation (Mountain View, CA)
Application Number:08/952,980
Patent Claims:1. A method of making an electrotransport device for delivering a polypeptide agent through a body surface by electrotransport, said method comprising:

(a) providing a first electrode;

(b) providing a second electrode;

(c) providing a power source electrically connected to said first and said second electrodes;

(d) providing at least one donor reservoir associated with an electrode; and

(e) providing a therapeutically effective amount of the polypeptide agent wherein the polypeptide agent is an analog of a parent polypeptide wherein said parent polypeptide comprises an .alpha.-helix and/or a .beta.-sheet segment, the analog polypeptide having one or more amino acid residues substituted relative to said parent polypeptide with an amino acid residue selected from the group consisting of Pro, Gly and Asn to disrupt said .alpha.-helical and/or .beta.-sheet segment; and

(f) adding said polypeptide agent to said donor reservoir to result in an electrotransport device for delivering said polypeptide agent through said body surface.

2. The method of claim 1, wherein said parent polypeptide is hirulog as depicted in FIG. 2A (SEQ ID NO:10).

3. The method of claim 2, wherein said polypeptide analog is hirulog-B2 as depicted in FIG. 2C (SEQ ID NO:12).

4. The method of claim 1, wherein said parent polypeptide is parathyroid hormone (PTH) as depicted in FIG. 1A (SEQ ID NO:6).

5. The method of claim 1, wherein said analog is selected from the group consisting of a PTH analog as depicted in FIG. 1C (SEQ ID NO:8) and FIG. 1D (SEQ ID NO:9).
