Title: Molecular methods of hybrid seed production
Abstract:A process is described for producing fertile hybrid seed or hybrid seed comprising fertile and sterile seed using male-sterile plants created by employing molecular techniques to manipulate anti-sense gene and other genes that are capable of controlling the production of fertile pollen in plants. Said plants are functionally male-sterile plants with pollen from male-fertile plants. Hybrid seed production is simplified and improved by this invention and can be extended to plant crop species for which commercially acceptable hybrid seed production methods are not currently available.

Patent Claims:1. An isolated recombinant DNA molecule comprising:

(a) a DNA sequence which encodes an antisense RNA which is antisense to a sense gene essential to pollen formation or function; and

(b) a promoter capable of causing the transcription of said DNA sequence.

2. The recombinant DNA molecule of claim 1, wherein said antisense gene comprises a DNA sequence which is antisense to any one of the genes in clones L4, L10, L16 or L19.

3. The recombinant DNA molecule of claim 1, wherein said DNA sequence is selected from a group consisting of base pairs 1-8585 of clone L4, the transcribed region of gene Bp4A of clone L4 comprising nucleotides 237-1427, the first coding region of gene Bp4A of clone L4 comprising nucleotides 308-368, the second coding region of gene Bp4A of clone L4 comprising nucleotides 1133-1261, the intron region of Bp4A comprising nucleotides 369-1132, the transcribed region of gene Bp4C of clone L4 comprising nucleotides 6296-7490, the first coding region of gene Bp4C of clone L4 comprising nucleotides 6369-6429, the second coding region of gene Bp4C of clone L4 comprising nucleotides 7198-7353, and the intron region of Bp4C comprising nucleotides 6430-7197.

The recombinant DNA molecule of claim 1, wherein said DNA sequence is selected from the group consisting of base pairs -789-2852 of the isolated nucleotide sequence of clone L10, the transcribed region of clone L10 comprising nucleotides 1-2107, the first coding region of clone 10 comprising nucleotides 75-315, the second coding region of clone L10 comprising nucleotides 475-1585, the third coding region of clone L10 comprising nucleotides 1673-1988, the first intron region of clone L10 comprising nucleotides 316-473, and the second intron region of clone L10 comprising nucleotides 1586-1672.

The recombinant DNA molecule of claim 1, wherein said DNA sequence is selected from the group consisting of base pairs of the nucleotide sequence of clone L19 comprising nucleotides -2022-2955, the transcribed region of L19 comprising nucleotides 1-2073, the first coding region of clone L19 comprising nucleotides 136-1201, the second coding region of clone L19 comprising nucleotides 1339-2024, and the intron region of clone L19 comprising nucleotides 1202-1338.

6. The recombinant DNA molecule of claim 1, wherein said DNA sequence comprises base pairs 1-384 of clone cBp401.

7. The recombinant DNA molecule of claim 1, wherein said DNA sequence comprises base pairs 1-364 of the gene in clone cBp405.

8. The recombinant DNA molecule of claim 1, wherein said DNA sequence comprises base pairs 1-298 of the gene in clone cBp408.

9. A plant that has been transformed with a recombinant DNA molecule according to claim 1, wherein said promoter is an inducible promoter which functions in said plant to cause transcription of said DNA sequence into RNA at about the time of transcription of said sense gene.

A method of producing hybrid plant seed by crossing a genetically transformed male-sterile female parent plant with a suitable male-fertile male parent plant, said genetically transformed male-sterile female parent plant containing one or more recombinant DNA molecules according to claim 1, said suitable male-fertile male parent plant containing a recombinant DNA sequence which compensates for the gene function that has been compromised or negates the disruption caused to said tissues or cells critical to pollen formation or function in said genetically transformed male-sterile female parent plant.

11. A method of producing hybrid plant seed comprising the steps of:

(a) producing a genetically transformed plant by:

(i) obtaining a transformed plant cell transformed with the recombinant DNA molecule according to claim 1; and

(ii) regenerating from said transformed plant cell a plant which is genetically transformed and which is male sterile or carries the male sterile trait;

(b) increasing the number of genetically transformed plants by:

(i) fertilizing the genetically transformed plant described in step (a) with pollen produced by a suitable male-fertile male plant, in a manner to effect a seed increase of genetically transformed plants; or

(ii) propagating said genetically transformed plant described in step (a); and

(c) effecting a hybrid cross by pollinating said genetically transformed plants with pollen from suitable male fertile plant donors.

12. A method of producing hybrid plant seed comprising the steps of:

(a) inserting into the genome of one or more plant cells of said plant the recombinant DNA molecule of claim 1;

(b) selecting a plant cell into which the recombinant DNA molecule is stably integrated;

(c) regenerating from the selected plant cell a plant which is male sterile;

(d) increasing the number of male sterile plants to produce a male sterile line by clonal propagation or by crossing the male sterile plant with a suitable male fertile plant and selecting plants which express said recombinant DNA molecules and repeating the steps of crossing and selecting as required using progeny of such cross(es) which have been selected; and

(e) crossing said male sterile line with a suitable male fertile plant line without said recombinant DNA molecule to obtain hybrid seed, said male fertile plant line is selected such that the hybrid seed is capable of growing into a male fertile plant, most plants of said male fertile plant line have at least one restorer DNA molecule encoding a restorer gene product integrated into their genome, said restorer DNA molecule or said restorer gene product being capable of restoring the function or development of a cells or tissues of a plant that is essential to pollen formation or function that is selectively interfered with by said gene recombinant DNA molecule.

13. Hybrid seed of a pollen-producing plant having a nuclear genome incorporating one or more recombinant DNA molecules as claimed in claim 1, and at least one restorer DNA molecule which is capable of restoring the function of the sense gene.

14. A method of producing a male sterile plant comprising the steps of:

(a) obtaining a transformed plant cell that has been genetically transformed with a DNA molecule according to claim 1; and

(b) regenerating from said transformed plant cell a genetically transformed plant.

15. The method according to claim 14, wherein the promoter is a constitutive promoter.

16. The method according to claim 14, wherein the promoter is an inducible promoter.
