Title: Aminoterminal propeptides of typed 1 procollagen
Abstract:Aminoterminal propeptide of type I procollagen exists in serum in two forms: classical type I procollagen which is a heterotrimer containing two pro .alpha.1-chains and one pro .alpha.2-chain of type I procollagen, and an .alpha.1-homotrimer type I procollagen containing three identical pro .alpha.1-chains. These intact, trimeric aminoterminal propeptides may be isolated without the use of proteolytic enzymes and the resultant propeptides may be used to prepare antibodies specific for the intact trimeric propeptide, having no affinity for the monomeric form of the propeptide. Such antibodies are useful in methods of assaying intact trimeric aminoterminal propeptide of type I procollagen in serum, without false information resulting from inadvertent assay of the monomeric form of the propeptide.

Patent Claims:1. An isolated intact trimeric .alpha.-1 homotrimer aminoterminal propeptide of type I procollagen wherein said propeptide is isolated from a biological sample under nondenaturing conditions and without the use of proteolytic enzymes.

2. An isolated intact heterotrimeric aminoterminal propeptide of type I procollagen wherein said propeptide is isolated from a biological sample under nondenaturing conditions and without the use of proteolytic enzymes.

3. A propeptide as claimed in claim 1 wherein said enzyme is capable of degrading the propeptide to its monomeric form.

4. A propeptide as claimed in claim 2 wherein said enzyme is capable of degrading the propeptide to its monomeric form.

5. A propeptide of claim 1 .alpha.1-homotrimeric which is free from bacterial collagenase.

6. A propeptide of claim 2 an isolated which is free from bacterial collagenase.

7. A propeptide according to any one of claim 1, 2, 3, 4, 5 or 6 wherein a label is bound to the propeptide.

8. A propeptide according to claim 7 wherein the label is selected from the group consisting of a radioactive label, a lanthanide, an enzyme, a luminescent label and a fluorescent label.

9. A propeptide according to claim 7 wherein the label is a radioactive label.
