Abstract:Synthetic polypeptide analogs of parathyroid hormone PTH, parathyroid hormone related peptide PTHrp, and of the physiologically active truncated homologs and analogs of PTH and PTHrp, in which amino acid residues (22-31) form an amphipathic .alpha.-helix, said residues (22-31) selected from hydrophilic amino acids (Haa) and lipophilic amino acids (Laa) ordered in the sequence: and their pharmaceutically acceptable salts are useful for the prophylaxis and treatment of corticosteroid induced osteopenia in mammals.

Patent Claims:1. A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitutions are selected from the group consisting of SEQ ID Nos. (26, 27, 28, 29, 30, 85 and 86).

2. A method of claim 1 in which the effective amount of polypeptide is from about 0.002 .mu.g of polypeptide/kg of patient body mass/day to about 10 .mu.g of polypeptide/kg of patient body mass/day.

3. A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified parathyroid hormone (PTH) or parathyroid hormone related peptide (PTHrP) having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 26.

A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 27.

A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 28.

A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 29.

7. A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 30.

8. A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 85.

9. A method for treating corticosteroid induced osteopenia, which method comprises administering to a subject in need thereof an effective amount of a modified PTH or PTHrP having bone mass restoring activity which differs from naturally occurring PTH or PTHrP by changes comprising substitutions at one or more of positions (22-31), wherein the substitution consists of SEQ ID No. 86.
