Title: Purified mite allergen
Abstract:The present invention is directed to novel purified and isolated mite allergens possessing mite allergen activity with a molecular weight of about 94,000, about 40,000, about 16,000 or about 14,000 as determined by SDS-PAGE, which can be isolated from extracts of mite, and to a process for producing such mite allergens. The purified and isolated mites allergens of the present invention are useful as a pharmaceutical and a diagnostic composition for mite allergic diseases.

Patent Claims:1. A purified and isolated mite allergen, said allergen possessing the following physicochemical and biological properties:

1 being isolated and purified from extracts of Dermatophagoides farinae mite bodies;

2 being a glycoprotein containing about 60% sugar;

3 having a molecular weight of about 94,000 as determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis;

4 exhibiting a single precipitation band with rabbit anti-mite serum as immunoassayed by cross immunoelectrophoresis;

possessing allergen activity; and

6 exhibiting mobility on sodium dodecyl sulfate-polyacrylamide gel electrophoresis which is the same under reducing and non-reducing conditions.

2. The purified and isolated mite allergen of claim 1, wherein said allergen has the following amino acid composition, neutral sugar composition and amino sugar composition:

Amino acid composition, of 3260.1 nmol/mg:

Asp 413.2, Glu 521.4, Cys 21.5, Ile 77.0, Phe 55.7, Arg 68.3, Thr 461.0, Glv 127.8, Val 212.5, Leu 370.4, His 30.3, Pro 107.7, Ser 298.0, Ala 150.4, Met 19.4, Tvr 3.3, Lys 322.2;

Neutral sugar composition, of 3154 nmol/mg:

Fuc 216.5, Gal 350.0, Ara 426.7, Man 1671.7, Xyl 490.0 and substantially free of glucose;

Amino sugar composition, of 265.2 nmol/mg:

HexAc 265.2.

3. The purified and isolated mite allergen of claim 1, wherein said allergen:

is freely soluble in water;

has a molecular weight of about 94,000 daltons as determined by sodium dodecyl sulfate=polyacrylamide gel electrophoresis;

has the following amino acid composition and sugar composition:

Amino acid composition, of 3260.1 nmol/mg:

Asp 413.2, Glu 521.4, Cys 21.5, Ile 77.0, Phe 55.7, Arg 68.3, Thr 461.0, Gly 127.8, Val 212.5, Leu 370.4, His 30.3, Pro 107.7, Ser 298.0, Ala 150.4, Met 19.4, Tyr 3.3, Lys 322.2;

Neutral sugar composition, of 3154 nmol/mg:

Fuc 216.5, Gal 350.0, Ara 426.7, Man 1671.7, Xyl 490.0 and substantially free of glucose;

Amino sugar composition, of 265.2 nmol/mg:

HexAc 265.2.

4. A purified and isolated mite allergen produced by a process comprising:

(a) growing Dermatophagoides farinae in a suitable medium at a temperature of 26.+-.2.degree. C. and 75% relative humidity until the mite density reaches 20,000 to 30,000 mites/gram of medium;

(b) killing said mites by freezing;

(c) adding a 10-fold amount by weight of a saturated sodium chloride solution to the mite culture and then stirring to produce a mixture;

(d) allowing said mixture to stand for one hour at 4.degree. C.;

(e) centrifuging said mixture at 3,000 rpm for 30 minutes at 4.degree. C. to suspend said mites on the surface of the supernatant solution;

(f) recovering said mites by suction;

(g) adding a half amount by weight of phosphate buffered saline to said mites to produce a mixture;

(h) grinding said mixture;

(i) adding additional phosphate buffered saline to the around mixture of step (h) to reach a 2-fold final amount, followed by one hour of extraction with occasional stirring;

(j) centrifuging the mixture of step (i) at 8,000 rpm for 30 minutes at 4.degree. C. to produce a supernatant;

(k) dialyzing said supernatant of step (j) against deionized water;

(l) removing insoluble substances by centrifugation at 8,000 rpm for 30 minutes at 4.degree. C.;

(m) fractionating the solution of step (1) by gel filtration chromatography through ULTROGEL AcA54 to produce a fraction exhibiting high skin reaction activity in asthmatic patients and high allergen activity in a histamine release test;

(n) fractionating said fraction of step (m) by preparative disc sodium dodecyl sulfate-polyacrylamide gel electrophoresis to produce a protein having a molecular weight of 94 kD; and

(o) recovering said 94 kD protein.

5. A pharmaceutical composition for the treatment of mite allergic diseases, which comprises as the active ingredient a pharmaceutically effective amount of the purified and isolated mite allergen of any one of claims 1, 2, 3 or 4, and a pharmaceutically acceptable carrier or diluent.

6. A method of treatment for mite allergic diseases which comprises administering a pharmaceutically effective amount of said purified and isolated mite allergen of claim 2.

7. The method of treatment for mite allergic diseases of claim 6, which is a hyposensitization therapy.

8. The method of claim 7, wherein said mite allergen is used in formulation with an adjuvant and at least one additive selected from the group consisting of a stabilizer, an excipient, a dissolution aid, an emulsifier, a buffer, an analgesic, a preservative, and a colorant.

9. The method of claim 7, wherein said mite allergen is administered per os, intracutaneously, subcutaneously, intramuscularly, or intraperitoneally.

10. The method of claim 7, wherein said mite allergen is used in the form of a troche, a sublingual tablet, an ophthalmic solution, an intranasal spray, a poultice, a cream, or a lotion.

11. The method of claim 7, wherein said mite allergen is administered in an amount of less than about 20 .mu.g once per week.
