Summary for Patent: 10,335,436
Title:Medical treatment composition comprising mammalian dental pulp stem cells
Abstract: A medical implant comprising in components from a tooth and stem cells harvested from at least one tooth. Pluripotent stem cells, other cells, and biologic constituents of the dental pulp may be harvested from the dental pulp of mammalian teeth, such as unerupted third molars in humans. After the stem cells are removed and isolated from the other teeth tissue, the hard tooth may be ground into a base material for the manufacture of a porous matrix into which the pluripotent stem cells, other cells, and biologic constituents of the dental pulp can be added. Additionally, soft tissue from the harvested tooth may be used as a carrier scaffold for soft tissue applications such as meniscal or cartilage repair.

Patent Claims:1. A composition comprising: a biological material obtained from a cryopreserved mammalian dental pulp, the cryopreserved mammalian dental pulp including a cryoprotectant being dimethyl sulfoxide, wherein the biological material is in suspension form and includes: (i) a plurality of non-cultured stem cells, including hematopoietic stem cells, and (ii) extracellular matrix proteins; wherein the composition is in a form that is safe for injection into a subject to treat a disease or injury.

2. The composition of claim 1, wherein the stem cells and extracellular matrix proteins are derived from mammalian dental pulp.

3. The composition of claim 2, wherein the dental pulp is equine dental pulp.

4. The composition of claim 3, wherein the equine dental pulp is chosen from neonatal equine dental pulp and fetal equine dental pulp.

5. The composition of claim 3, wherein the equine dental pulp is taken from the premolar or molar teeth of the mandibular and maxillary regions.

6. The composition of claim 1, wherein the composition is in a form chosen from a single cell suspension and a multiple cell suspension.

7. The composition of claim 1 wherein the stem cells are chosen from pluripotent mammalian dental pulp stem cells, multipotent mammalian dental pulp stem cells, and mixtures thereof.

8. The composition of claim 1, further comprising one or more growth factors.

9. The composition of claim 8, wherein the one or more growth factors are chosen from transforming growth factor beta, insulin like growth factor I, insulin like growth factor II, platelet-derived growth factor, fibroblast growth factor, and bone morphogenetic proteins.

10. The composition of claim 1, wherein the stem cells include CD34 positive, CD44 positive, CD45 positive, CD90 positive, and CD105 positive cells.

11. The composition of claim 1, further comprising the hard components of a tooth.

12. The composition of claim 11, wherein the hard components of the tooth are chosen from dentin and enamel.

13. The composition of claim 11, wherein the hard components of the tooth have been processed into a particulate, ground, or powder form.

14. The composition of claim 1, further comprising hydroxyapatite.

15. The composition of claim 1, wherein the extracellular matrix proteins include at least one protein chosen from an anti-inflammatory protein, an anti-degradative protein, interleukin-1 receptor antagonist, collagen type I, and glycosaminoglycans.

16. The composition of claim 1, further comprising a vehicle for the stem cells and extracellular matrix proteins, the vehicle comprising a balanced salt and isotonic solution.

17. The composition of claim 1, wherein the stem cells and extracellular matrix proteins are derived from an equine source.

18. The composition of claim 1, wherein said extracellular matrix proteins include collagen.

19. The composition of claim 1, further comprising blood concentrates or tissue derivatives from an autologous source.

20. The composition of claim 1, further comprising blood concentrates or tissue derivatives from an allogenous source.

21. The composition of claim 1, further comprising a portion of a soft tissue component from the mammalian tooth papilla.

22. The composition of claim 1, wherein said composition is a component of a medical device.

23. The composition of claim 1, wherein the biological material is a digestate from dental pulp.

24. The composition of claim 1, wherein the plurality of stem cells is divided into subpopulations of stem cells.

25. The composition of claim 24, wherein the subpopulations include at least a subpopulation of hematopoietic stem cells and a subpopulation of mesenchymal stem cells.

26. A composition comprising: a biological material obtained from a filtered mammalian dental pulp in particulate form, the mammalian dental pulp including a non-naturally occurring cryoprotectant, wherein the biological material is in suspension form and includes a plurality of stem cells and attached extracellular matrix proteins; wherein the composition is in a form that is suitable safe for injection into a subject to treat a disease or injury.

27. A composition comprising: a biological material obtained from a ground mammalian dental pulp, the mammalian dental pulp including a non-naturally occurring cryoprotectant, wherein the biological material is in suspension form and includes: (i) a plurality of stem cells, including hematopoietic stem cells, and (ii) extracellular matrix proteins; wherein the biological material has not been digested by collagenase; and wherein the composition is in a form that is suitable safe for injection into a subject to treat a disease or injury.
